Investigating the Mechanism of Sex-Dependent Effects of Methamphetamine on Dopamine Transmission in Vivo

Methamphetamine abuse is on the rise, yet the effects of sex differences on the activity of midbrain dopamine neurons after methamphetamine exposure remain unclear. Methamphetamine has been shown to increases the firing rate of dopaminergic neurons and alters network dynamics; however, the impact of chronic exposure on dopamine neurons in males and females is still unknown. We hypothesize that methamphetamine exposure affects dopamine neuron activity are increased in females compared to males. We will test this hypothesis by using conditioned place preference (CPP), a model of drug addiction. For these studies, we used in vivo imaging in freely behaving mice which enables us to correlate the behavioral responses and dopamine activity. My specific role to track behavioral differences (speed and total distance traveled) in male and female mice after repeated methamphetamine exposure. This research provides valuable insights into the neurobiological and neurobehavioral effects of sex-dependent differences in methamphetamine abuse.
Contrasting the Effect of
α-Synuclein Overexpression on Dopamine Transmission and Neural Viability in the SNc vs. the VTA

The synaptic protein α-Synuclein (αSyn) plays a key role in neurodegenerative diseases like Parkinson’s, but its impact on dopamine-producing neurons in the brain is not fully understood. Notably, dopamine neurons in the substantia nigra pars compacta (SNc) are more vulnerable to damage compared to those in the ventral tegmental area (VTA), though the reasons for this difference remain unclear. Understanding how αSyn overproduction differentially affects these regions is important for developing targeted treatments. My project focuses on understanding how αSyn affects dopamine transmission and neuronal viability in these different regions. We hypothesize αSyn overexpression reduces dopamine synthesis in the SNc prior to its reduction in the VTA. To explore this, we utilized two animal models that overproduce αSyn in SNc and VTA. I will perform immunolabeling to measure αSyn and tyrosine hydroxylase expression to assess the health of dopamine-producing neurons in these regions. With our study, we aim to deepen the understanding of how αSyn affects different brain regions in the early stages of Parkinson’s disease, providing insights that could guide future treatments.

My involvement in the UF MARC program has given me invaluable research experience and academic support as an undergraduate student. Volunteering with Project Downtown Gainesville has allowed me to contribute to the well-being of the local community by providing essential services to homeless and disadvantaged individuals. This experience has been incredibly rewarding, offering the chance to build lasting relationships with those we serve and make a positive impact on the lives of others. Through consistent support, I've been able to engage in meaningful humanitarian work that directly benefits our community.
